Microwave-Enabled Fast Preparation of a Metal-Organic Framework Hybrid Membrane for Filtration-Enhanced Simultaneous Separation and Detection of Aflatoxin B1.

Mycotoxin contamination in food and the environment seriously harms human health. Sensitive and timely detection of mycotoxins is crucial. Here, we report a dual-functional hybrid membrane with absorptivity and responsiveness for fluorescent-quantitative detection of mycotoxin aflatoxin B1 (AFB1). A biomineralization-inspired and microwave-accelerated fabrication method was established to prepare a hybrid membrane with a metal-organic framework (MOF) loaded in high density. The MOF presented high efficiency in capturing AFB1 and showed fluorescence intensity alteration simultaneously, enabling a dual adsorption-response mode. Deriving from the inherent porous structure of the hybrid membrane and the absorptive/responsive ability of the loaded MOF, a filtration-enhanced detection mode was elaborated to provide a 1.67-fold signal increase compared with the conventional soaking method. Therefore, the hybrid membrane exhibited a rapid response time of 10 min and a low detection limit of 0.757 ng mL-1, superior to most analogues in rapidity and sensitivity. The hybrid membrane also presented superior specificity, reproducibility, and anti-interference ability and even performed well in extreme environments such as strong acid or alkaline, satisfying the practical requirements for facile and in-field detection. Therefore, the membrane had strong applicability in chicken feed samples, with a detection recovery between 70.6% and 101%. The hybrid membrane should have significant prospects in the rapid and in-field inspection of mycotoxins for agriculture and food.

Emergent Alternaria toxins present in naturally contaminated dried tomatoes and their reduction using cold atmospheric plasma

This study investigated the efficacy of cold atmospheric plasma (CAP) treatment in degrading the emerging Alternaria toxins as pure molecules and as natural contaminants in dried tomatoes. The analyses of 25 batches of dried tomatoes showed that more than 40% of samples were contaminated with tenuazonic acid (TeA), one with Alternariol (AOH), while Tentoxin (TEN), Alternariol monomethyl ether (AME) and Altenuene (ALT) were not detected. CAP under ozone (O3) and NOx regimes were used to reduce the mycotoxin contamination, the results shown that O3 regime was more effective against all mycotoxins tested, while NOx regime showed different efficacy depending on the specific mycotoxin. The Weibull model proved to be appropriate for the degradation kinetics of all Alternaria toxins, regardless of the kinetic order they possess. Under the O3 regime, the resistance of the mycotoxin was AME> AOH > TEN > TeA > ALT. Our studies have shown for the first time that CAP reduces Alternaria toxins in naturally contaminated dried tomatoes. In particular, the rate of degradation under O3 regime increased significantly with the longest exposure time, reaching 52.80±1.21 % of mycotoxin degradation. Our results expand our knowledge of CAP treatment as a potential means of reducing mycotoxin contamination in food. Further studies will be carried out to investigate the potential mycotoxin degradation products and to ensure food safety.

Development of a microfluidic device to enrich and detect zearalenone in food using quantum dot-embedded molecularly imprinted polymers.

Mycotoxins are secondary metabolites of certain moulds, prevalent in 60-80% of food crops and many processed products but challenging to eliminate. Consuming mycotoxin-contaminated food and feed can lead to various adverse effects on humans and livestock. Therefore, testing mycotoxin residue levels is critical to ensure food safety. Gold standard analytical methods rely on liquid chromatography coupled with optical detectors or mass spectrometers, which are high-cost with limited capacity. This study reported the successful development of a microfluidic "lab-on-a-chip" device to enrich and detect zearalenone in food samples based on the fluorescence quenching effect of quantum dots and selective affinity of molecularly imprinted polymers (MIPs). The dummy template and functional polymer were synthesized and characterized, and the detailed microfluidic chip design and optimization of the flow conditions in the enrichment module were discussed. The device achieved an enrichment factor of 9.6 (±0.5) in 10 min to quantify zearalenone spiked in food with high recoveries (91-105%) at 1-10 mg kg-1, covering the concerned residue levels in the regulations. Each sample-to-answer test took only 20 min, involving 3 min of manual operation and no advanced equipment. This microfluidic device was mostly reusable, with a replaceable detection module compatible with fluorescence measurement using a handheld fluorometer. To our best knowledge, the reported device was the first application of an MIP-based microfluidic sensor for detecting mycotoxin in real food samples, providing a novel, rapid, portable, and cost-effective tool for monitoring mycotoxin contamination for food safety and security.

Stakeholder assessment for mycotoxin analysis: exploring the demand along the European food supply chain

Abstract Mycotoxins are amongst the most prevalent food contaminants leading to serious health implications for humans and animals. Limiting exposure to them within the population remains a pressing food safety challenge. Prevention and timely detection are crucial for minimising mycotoxin contamination in food and feed. Therefore easy-to-use, rapid, eco-friendly and low-cost screening methods are increasingly implemented. Early-stage end-user engagement in the technological development process aids and guides the research towards increased societal impact. To investigate these end-user needs, the limitations of the currently used methods and the expectations towards a novel approach were mapped among stakeholders in a comprehensive survey. Stakeholders along the food supply chain (agricultural, food processing, retail, food safety and control, laboratories) were identified in five European countries and contacted. A total of 63 participants completed the survey, which was followed-up by an interview. The results of the survey revealed that different end-user groups have different priorities. Important limiting factors for agricultural, processing and retail stakeholders of the current methodologies include the complexity of sample preparation, high cost and time-to-results. Complementing the accredited laboratory tests with a pre-screening device would be especially interesting for agricultural producers (87.5% of the participants are interested) and food processors (80%), since there is an increasing demand for on-site detection of contamination. On the other hand, food control authorities and commercial laboratories indicated higher priority towards low quantification limits and multi-compound methods. The time to get the results was found to be more important than the testing cost (important aspect for 74.6 versus 66.7%). Overall, the findings of this study are critical input for end-user-targeted development of novel mycotoxin detection platforms.

Detoxification Impacts of Dietary Probiotic and Prebiotic Supplements Against Aflatoxins: An Updated Knowledge – A Review

Abstract The widespread prevalence of food pollutants seriously threatens human and animal health. Mycotoxins are secondary metabolites primarily formed by toxigenic fungal genera, including Aspergillus, Penicillium, Fusarium, and Alternaria, demonstrating one of the principal pollutants in diets or feed products. Mycotoxin contamination in food can harm health, including stunted development, immune system suppression, infertility, vomiting, and gastrointestinal and cancerous conditions. These effects can occur both acutely and chronically. The complex food chain can be contaminated with mycotoxins at any point, including during harvest, industrial processing, shipping, or storage, putting the food sector under societal pressure owing to the waste generated by infected goods. One of the biological controls of mycotoxin is provided by probiotics and prebiotics, controlled as foods and dietary supplements made of bacteria or yeast. Aflatoxin’s bioavailability and gastrointestinal absorption can be reduced using various probiotics and prebiotics.

DNA Damage Induced by T-2 Mycotoxin in Human Skin Fibroblast Cell Line-Hs68.

T-2 mycotoxin is the most potent representative of the trichothecene group A and is produced by various Fusarium species, including F. sporotrichioides, F. poae, and F. acuminatum. T-2 toxin has been reported to have toxic effects on various tissues and organs, and humans and animals alike suffer a variety of pathological conditions after consumption of mycotoxin-contaminated food. The T-2 toxin's unique feature is dermal toxicity, characterized by skin inflammation. In this in vitro study, we investigated the molecular mechanism of T-2 toxin-induced genotoxicity in the human skin fibroblast-Hs68 cell line. For the purpose of investigation, the cells were treated with T-2 toxin in 0.1, 1, and 10 μM concentrations and incubated for 24 h and 48 h. Nuclear DNA (nDNA) is found within the nucleus of eukaryotic cells and has a double-helix structure. nDNA encodes the primary structure of proteins, consisting of the basic amino acid sequence. The alkaline comet assay results showed that T-2 toxin induces DNA alkali-labile sites. The DNA strand breaks in cells, and the DNA damage level is correlated with the increasing concentration and time of exposure to T-2 toxin. The evaluation of nDNA damage revealed that exposure to toxin resulted in an increasing lesion frequency in Hs68 cells with HPRT1 and TP53 genes. Further analyses were focused on mRNA expression changes in two groups of genes involved in the inflammatory and repair processes. The level of mRNA increased for all examined inflammatory genes (TNF, INFG, IL1A, and IL1B). In the second group of genes related to the repair process, changes in expression induced by toxin in genes-LIG3 and APEX were observed. The level of mRNA for LIG3 decreased, while that for APEX increased. In the case of LIG1, FEN, and XRCC1, no changes in mRNA level between the control and T-2 toxin probes were observed. In conclusion, the results of this study indicate that T-2 toxin shows genotoxic effects on Hs68 cells, and the molecular mechanism of this toxic effect is related to nDNA damage.

Antifungal and Antiaflatoxigenic Activities of Massoia Essential Oil and C10 Massoia Lactone against Aflatoxin-Producing Aspergillus flavus.

Fungal infection and mycotoxin contamination are major hazards to the safe storage and distribution of foods and feeds consumed by humans and livestock. This study investigated the antifungal and antiaflatoxigenic activities of massoia essential oil (MEO) and its major constituent, C10 massoia lactone (C10), against aflatoxin B (AFB)-producing Aspergillus flavus ATCC 22546. Their antifungal activities were evaluated using a disc diffusion assay, agar dilution method, and a mycelial growth inhibition assay with the AFB analysis using liquid chromatography triple quadrupole mass spectrometry. MEO and C10 exhibited similar antifungal and antiaflatoxigenic activities against A. flavus. C10 was a primary constituent in MEO and represented up to 45.1% of total peak areas analyzed by gas chromatography-mass spectrometry, indicating that C10 is a major compound contributing to the antifungal and antiaflatoxigenic activities of MEO. Interestingly, these two materials increased AFB production in A. flavus by upregulating the expression of most genes related to AFB biosynthesis by 3- to 60-fold. Overall, MEO and C10 could be suitable candidates as natural preservatives to control fungal infection and mycotoxin contamination in foods and feeds as Generally Recognized As Safe (GRAS) in the Flavor and Extract Manufacturers Association of the United States (FEMA), and MEO is a more suitable substance than C10 because of its wider range of uses and higher allowed concentration than C10.

3-Acetyldeoxynivalenol induces apoptosis, barrier dysfunction and endoplasmic reticulum stress by inhibiting mTORC1-dependent autophagy in porcine enterocytes

3-Acetyldeoxynivalenol (3-Ac-DON), an acetylated form of deoxynivalenol, is widely present in mycotoxin-contaminated food, feed as well as in other natural sources. Ingestion of 3-Ac-DON may result in intestinal dysfunction, leading to gut diseases in humans and animals. Nevertheless, the molecular mechanism of 3-Ac-DON in intestinal epithelial cytotoxicity remains unclear. In this study, intestinal porcine epithelial cell line 1 (IPEC-1) cells were treated with different concentrations of 3-Ac-DON for 12 h or 24 h, respectively. The results showed that 3-Ac-DON caused decreased cell viability, cell cycle arrest in G1 phase and depolarization of mitochondrial membrane potential. Western blotting analysis showed that 3-Ac-DON significantly decreased the expression of tight junction proteins, inhibited autophagy and activated endoplasmic reticulum (ER) stress in IPEC-1 cells (P < 0.05). Further investigation demonstrated that 3-Ac-DON caused apoptosis, ER stress and barrier dysfunction were reversed after co-treatment with the autophagy activator rapamycin (100 nM), indicating that autophagy plays a key role in the process of 3-Ac-DON-induced cell damage. In addition, we demonstrated that 3-Ac-DON inhibits the occurrence of autophagy mediated by mTORC1 protein. In conclusion, our research indicated that the mTORC1 protein and autophagy played a key role in the 3-Ac-DON-induced cytotoxic in IPEC-1 cells, which would provide new therapeutic targets and ideas for 3-Ac-DON-mediated intestinal injury.

GC-ToF-MS Profiling and In Vitro Inhibitory Effects of Selected South African Plants against Important Mycotoxigenic Phytopathogens.

The harmful effects following the ingestion of mycotoxin-contaminated food include the induction of cancers, mutagenicity, immune suppression, and toxicities that target organs of the digestive, cardiovascular, and central nervous systems. Synthetic fungicides are generally associated with a high toxic residue in food and the development of excessive fungal resistance. This study aimed to determine the antifungal activities against mycotoxigenic fungi of selected South African plant leaves and potentially develop plant-derived bio-fungicides, and, furthermore, to explore the in vitro antioxidant activity and the phytochemical spectra of the compounds of the selected medicinal plant extracts. The extracts were tested for antifungal activity against phytopathogenic strains using a microdilution broth assay. Bauhinia galpinii extracts exhibited the lowest minimum inhibitory concentration (MIC) against C. cladospoides and P. haloterans at 24 h incubation periods. C. caffrum had good antioxidant activity against 2,2-diphenyl-1-picrylhydrazyl (DPPH) with 50% inhibitory concentration (IC50) values of 0.013 mg/mL while B. galpini had IC50 values of 0.053 against free radicals of 2,2'-azinobis (3-ethylbenzthiazoline-6-suphonic acid (ABTS). The antimycotoxigenic and antioxidant activity exerted by both B. galpinii and C. caffrum may well be attributed to high TPC. In the GC-ToF-MS analysis, all the selected medicinal plants exhibited the presence of Hexadecanoic acid at varying % areas, while both B. galpinii and C. caffum exhibited the presence of lupeol at % area 2.99 and 3.96, respectively. The compounds identified, particularly the ones with higher % area, may well explain the biological activity observed. Although the selected medicinal plants exhibited a notable biological activity, there is a need to explore the safety profiles of these plants, both in vitro and in vivo.

A MAPK signaling cascade regulates the fusaric acid-induced cell death in Arabidopsis

Mycotoxin contamination of foods and feeds is a global problem. Fusaric acid (FA) is a mycotoxin produced by Fusarium species that are phytopathogens of many economically important plant species. FA can cause programmed cell death (PCD) in several plant species. However, the signaling mechanisms of FA-induced cell death in plants are largely unknown. Here we showed that FA induced cell death in the model plant Arabidopsis thaliana, and MPK3/6 phosphorylation was triggered by FA in Arabidopsis. Both the acid nature and the radical of FA are required for its activity in inducing MPK3/6 activation and cell death. Expression of the constitutively active MKK5DD resulted in the activation of MPK3/6 and promoted the FA-induced cell death. Our work demonstrates that the MKK5-MPK3/6 cascade positively regulates FA-induced cell death in Arabidopsis and also provides insight into the mechanisms of how cell death is induced by FA in plants.

Detoxifying mycotoxins and antifungal properties of two rumen-derived Enterococcus species in artificially contaminated corn silages.

Mycotoxins contamination in food and feed has emerged as an issue of serious concern because they pose serious health risks to both humans and livestock. The study aimed to evaluate the effects of two rumen-derived Enterococcus spp. on fermentation and hygienic quality of artificially contaminated corn silages. The toxigenic fungal-infested (FI) and non-fungal infested (NFI) corn was harvested at 1/2 milk line stage and ensiled without additives (CON) or with Enterococcus faecalis (E) or Enterococcus faecium (M). The pH of FI silages was higher than that of NFI silages, the pH in NFI-M was lower than in NFI-CON. Inoculating E. faecium markedly increased lactic acid concentration compared to CON and E silages. Both E. faecium and E. faecalis decreased the deoxynivalenol (DON) and zearalenone (ZEN) concentrations compared with the CON for FI silages, while E. faecium was more effective in eliminating aflatoxin B1 (AFB1 ). The FI silage had higher bacterial and fungal Shannon indexes than NFI silages. The relative abundance (RA) of Aspergillus and Fusarium marked a decline from day 5 to day 90. Inoculating E. faecium and E. faecalis reduced the RA of Penicillium compared to CON. In vitro mycotoxins removal assay indicated that E. faecium was more effective in AFB1 detoxification while having lower detoxifying ZEN capacity than E. faecalis. Inoculating rumen-derived Enterococcus spp. isolates alleviated the negative effects of fungal infestation on the fermentation and hygienic quality of corn silages by changing the microbial communities and detoxifying mycotoxins. © 2023 Society of Chemical Industry.

Combined toxicity assessment of a naturally occurring toxin and a triazole fungicide on different biological processes through toxicogenomic data mining with mixtures

Fungicides are widely used to prevent fungal growth and reduce mycotoxin contamination in food, which provides the opportunity for the co-occurrence of mycotoxins and fungicide residues in food and poses a greater risk to human health. To assess the combined effects of a naturally occurring mycotoxin, citrinin (CIT), and a widely used triazole fungicide, triadimefon (TAD) on different biological processes, the comparative toxicogenomics database was used to obtain phenotypes and response genes for CIT or TAD exposure. Then individual and combined exposure models were developed with zebrafish embryos, and the interaction between CIT and TAD was analyzed using the 2 × 2 factorial design approach to observe the toxic effects. Through data mining analysis, our results showed that CIT or TAD exposure is related to different biological phenotypes, such as cell death, regulation of antioxidant systems, and thyroid hormone metabolism. Our results also showed that CIT (4-day LC50 value of 12.7 mg/L) exposure possessed higher toxicity to zebrafish embryos compared with TAD (4-day LC50 value of 29.6 mg/L). Meanwhile, individual exposure to CIT and TAD altered the expression levels of biomarkers related to oxidative stress, inflammation, apoptosis and hypothalamic-pituitary-thyroid (HPT) axis. Notably, combined exposure to CIT and TAD induced changes in the mentioned biological processes and had an interactive effect on the expression of multiple biomarkers. In conclusion, we evaluated the toxic effects of CIT and TAD in isolation and combination by in-vivo experiments, which provide a new methodological basis and reference for future risk assessment and setting of safety limits for foodborne toxicants.

Effect of Gliding Arc Plasma Jet on the Mycobiota and Deoxynivalenol Levels in Naturally Contaminated Barley Grains.

Fusarium graminearum and Fusarium meridionale are primary contaminants of barley, capable of producing several mycotoxins, mainly type B trichothecenes and zearalenone. Cold plasma decontamination has been gaining prominence, seeking to control the fungal and mycotoxin contamination of food and feed and to improve product quality. To reach this objective, the present study was divided into two parts. In the first part, F. meridionale and F. graminearum strains were exposed to gliding arc plasma jet (GAPJ). Cell viability tests showed the inactivation of F. meridionale after 15-min treatment, whereas F. graminearum showed to be resistant. In the second part, barley grains were treated by GAPJ for 10, 20, and 30 min, demonstrating a reduction of about 2 log CFU/g of the barley's mycobiota, composed of yeasts, strains belonging to the F. graminearum species complex, Alternaria, and Aspergillus. A decrease in DON levels (up to 89%) was observed after exposure for 20 min. However, an increase in the toxin Deoxynivalenol-3-glucoside (D3G) was observed in barley grains, indicating a conversion of DON to D3G.

Occurrence of Toxinogenic Fungi in Livestock Feeds in Eastern Algeria

Background: The ubiquitous nature of molds, their ability to colonize diverse substrates, and the lack of effective control measures have contributed to the high incidences of molds and mycotoxin contamination in foods and feeds. These dangerous secondary metabolites enter the food chain in the field, during storage, or later.&#x0D; Material and Methods: A total of 40 livestock feed samples were randomly collected from ten farms in eastern Algeria. The ten types of analyzed feed were: wheat bran, dried bread, alfalfa, straw, oat hay, barley, date scraps, grass silage, corn silage, and concentrate.&#x0D; Results and Discussion: Results show that all the samples were contaminated with different species of fungi. A total of 248 fungal strains were isolated. In terms of frequency, the genus Aspergillus was the most frequent fungi recovered from 32 samples (80%) between all feeds studied, followed by the genus Penicillium recovered from 31 samples (77.5%).&#x0D; Conclusion: As a preventive measure, appropriate agricultural practices could provide unfavorable conditions for the development of fungi and toxinogenesis. However, the installation of a mycotoxins-monitoring program may be necessary to protect animals' and consumers' health.

Current Status of Major Mycotoxins Contamination in Food and Feed in Asia─A Review

Mycotoxins can cause genotoxic, mutagenic, teratogenic, and carcinogenic effects in humans and animals. Mycotoxins inevitably exist widely in all kinds of food and feed commodities, of which cereals are the most susceptible. In one of the largest cereal producing and consuming regions in the world, mycotoxin contamination causes about 1 billion metric tons of food loss annually, which also influences a serious decline of Asian economic development. This Review is based on three databases, PubMed, Google Scholar, and Web of Science, including studies that were published in English in the period from January 1, 2016 to October 1, 2022. Results show the current status and control strategies of the aflatoxins, ochratoxin A, zearalenone, deoxynivalenol, and fumonisins in Asia. Developed countries in Asia should maintain strict monitoring and establish an effective control system for the whole food production chain, while developing countries should strengthen the prevention and control strategies for mycotoxins.

Genetic Regulation of Mycotoxin Biosynthesis.

Mycotoxin contamination in food poses health hazards to humans. Current methods of controlling mycotoxins still have limitations and more effective approaches are needed. During the past decades of years, variable environmental factors have been tested for their influence on mycotoxin production leading to elucidation of a complex regulatory network involved in mycotoxin biosynthesis. These regulators are putative targets for screening molecules that could inhibit mycotoxin synthesis. Here, we summarize the regulatory mechanisms of hierarchical regulators, including pathway-specific regulators, global regulators and epigenetic regulators, on the production of the most critical mycotoxins (aflatoxins, patulin, citrinin, trichothecenes and fumonisins). Future studies on regulation of mycotoxins will provide valuable knowledge for exploring novel methods to inhibit mycotoxin biosynthesis in a more efficient way.

Zearalenone disturbs the reproductive-immune axis in pigs: the role of gut microbial metabolites

BackgroundExposure to zearalenone (ZEN, a widespread Fusarium mycotoxin) causes reproductive toxicity and immunotoxicity in farm animals, and it then poses potential threats to human health through the food chain. A systematic understanding of underlying mechanisms on mycotoxin-induced toxicity is necessary for overcoming potential threats to farm animals and humans. The gastrointestinal tract is a first-line defense against harmful mycotoxins; however, it remains unknown whether mycotoxin (e.g., ZEN)-induced toxicity on the reproductive-immune axis is linked to altered gut microbial metabolites. In this study, using pigs (during the three phases) as an important large animal model, we investigated whether ZEN-induced toxicity on immune defense in the reproductive-immune axis was involved in altered gut microbial-derived metabolites. Moreover, we observed whether the regulation of gut microbial-derived metabolites through engineering ZEN-degrading enzymes counteracted ZEN-induced toxicity on the gut-reproductive-immune axis.ResultsHere, we showed ZEN exposure impaired immune defense in the reproductive-immune axis of pigs during phase 1/2. This impairment was accompanied by altered gut microbial-derived metabolites [e.g., decreased butyrate production, and increased lipopolysaccharides (LPS) production]. Reduction of butyrate production impaired the intestinal barrier via a GPR109A-dependent manner, and together with increased LPS in plasma then aggravated the systemic inflammation, thus directly and/or indirectly disturbing immune defense in the reproductive-immune axis. To validate these findings, we further generated recombinant Bacillus subtilis 168-expressing ZEN-degrading enzyme ZLHY-6 (the Bs-Z6 strain) as a tool to test the feasibility of enzymatic removal of ZEN from mycotoxin-contaminated food. Notably, modified gut microbial metabolites (e.g., butyrate, LPS) through the recombinant Bs-Z6 strain counteracted ZEN-induced toxicity on the intestinal barrier, thus enhancing immune defense in the reproductive-immune axis of pigs during phase-3. Also, butyrate supplementation restored ZEN-induced abnormalities in the porcine small intestinal epithelial cell.ConclusionsAltogether, these results highlight the role of gut microbial-derived metabolites in ZEN-induced toxicity on the gut-reproductive-immune axis. Importantly, targeting these gut microbial-derived metabolites opens a new window for novel preventative strategies or therapeutic interventions for mycotoxicosis associated to ZEN.

Recovery of Energy and Nutrients from Mycotoxin-Contaminated Food Products through Biological Treatments in a Circular Economy Perspective: A Review

Mycotoxins’ contamination of food products is a well-known issue that is gaining interest nowadays due to increasing contaminations that are also related to climate change. In this context, and considering the principles of Circular Economy, finding robust and reliable strategies for the decontamination and valorisation of mycotoxin-contaminated products becomes mandatory. Anaerobic digestion (AD) and composting appear as promising biological treatments to degrade mycotoxins and allow for recovering energy (i.e., biogas production) and materials (i.e., nutrients from digestate and/or compost). The aim of the present paper was to carry out an organic revision of the state of the art of energy and materials recovery from mycotoxin-contaminated food products through biological treatments, highlighting results and research gaps. Both processes considered were not generally affected by the contamination of the feedstocks, proving that these compounds do not affect process stability. Mycotoxins were highly removed due to the concurrence of microbiological and physical agents in AD and composting. From the literature review, emerged the points that still need to be addressed before considering large scale application of these processes, which are (i) to deepen the knowledge of biochemical transformations of mycotoxins during the processes, (ii) to assess the fate of mycotoxins’ residues and metabolites in soil once digestate/compost are applied, (iii) to evaluate and optimize the integration of AD and composting in order to increase the environmental and economical sustainability of the processes, and (iv) to update legislation and regulations to allow the agricultural reuse of organic fertilizers obtained from contaminated feedstocks.

Degradation of Alternaria mycotoxins by UV-C irradiation: Effect of selected process and exposure to food components

Alternariol (AOH) and alternariol monomethyl ether (AME) are two Alternaria mycotoxins with high occurrence rates in food systems. This study aimed to investigate the photodegradation of AOH and AME by ultraviolet-C (UV-C) irradiation. The effect of UV-C intensity, pH, treatment time, solvents and the exposure of food components were evaluated. After treated by UV-C irradiation at 3500 μW/cm2 for 90 min, AOH samples in methanol, aqueous solution and solid state were degraded by 89.1%, 72.9% and 53.2%, respectively, while the degradation percentages of AME were 86.6%, 50.1% and 11.1%, respectively. Increasing irradiation intensity and prolonging irradiation time could significantly facilitate the degradation of AOH and AME. An alkaline environment (pH = 11) was more conducive to the degradation of toxins. In addition, 2.5 mg mL−1 citric acid or malic acid increased the photodegradation of AOH and AME to 94.6% and 95.3%, 93.2% and 70.5%, respectively. However, protein, polyphenols and vitamin C exerted inhibitory effects on the degradation, while 10% glucose or sucrose reduced the photodegradation of AOH and AME to 65.9% and 40.3%. UV-C treatment could effectively reduce the content of AOH and AME, with the highest efficiency achieved in methanol and alkaline environment. By contrast, UV-C irradiation is more effective in degrading toxins in some liquid foods rich in organic acids but lacking in protein. The utilization of UV-C radiation appears to be a potentially useful approach for decreasing the underlying risk of Alternaria mycotoxin contamination in foods.

Toxicity and preventive approaches of Fusarium derived mycotoxins using lactic acid bacteria: state of the art.

Mycotoxin contamination of food and feed is a serious food safety issue and causes acute and chronic diseases in humans and livestock. Climatic and agronomic changes helps in the proliferation of fungal growth and mycotoxin production in food commodities. Mycotoxin contamination has attracted global attention due to its wide range of toxicity to humans and animals. However, physical and chemical management approaches in practice are unsafe for well-being due to their health-hazardous nature. Various antibiotics and preservatives are in use to reduce the microbial load and improve the shelf life of food products. In addition, the use of antibiotic growth promotors in livestock production may increase the risk of antimicrobial resistance, which is a global health concern. Due to their many uses, probiotics are helpful microbes that have a significant impact on food and nutrition. Furthermore, the probiotic potential of lactic acid bacteria (LAB) is employed in various food and feed preparations to neutralize mycotoxins, antimicrobial activities, balance the gut microbiome, and various immunomodulatory activities in both humans and livestock. In addition, LAB produces various antimicrobials, flavouring agents, peptides, and proteins linked to various food and health care applications. The LAB-based processes for mycotoxin management are more effective, eco-friendly, and low-cost than physical and chemical approaches. The toxicity, novel preventive measures, binding nature, and molecular mechanisms of mycotoxins' detoxification using LAB have been highlighted in this review.
